JCRB 細胞庫正在收集各種人類和動物培養細胞,包括癌症細胞和轉基因細胞。細胞資源分發給日本和世界各地的研究人員。這些細胞通過測試微生物污染、病毒污染和交叉培養污染進行了全面鑑定。一些細胞的特徵在於核型分析和/或細胞表面標記。我們與世界上其他主要細胞庫合作,開發細胞培養和質量控制方法,以支持醫藥科學的醫學基礎研究。
歷史
1985年
日本癌症研究資源銀行(JCRB)在現東京國立衛生科學研究所(NIHS)的前身國立衛生健康科學研究所成立。細胞庫基礎管理解決方案由ATCC引進。
1995年
JCRB更名為日本研究生物資源館。發行部門與 JCRB 分開,由日本健康科學基金會設立的健康科學研究資源銀行 (HSRRB) 啟動非免費分發。
2005年
國家生物醫學創新研究所(NIBIO)成立。 JCRB 細胞庫從 NIHS 轉移到 NIBIO,因此,從東京轉移到大阪茨城市。
2013 年
HSRRB 關閉,細胞系的分配由 JCRB 負責。
2015年

細胞系的品質控制
品質控制在細胞系的使用中非常重要,既考慮到結果的可重複性,又考慮到生物製品的安全性。從 JCRB 細胞庫分發的所有細胞系均已確認無污染,並透過以下品質控制進行驗證。從品質控制測試中獲得的數據包含在我們網站上每個細胞系的資訊中。根據要求為每個細胞系頒發分析證書。
1.細菌和真菌檢測
為了評估細胞系中細菌和真菌的污染,將細胞分散到三種針對細菌和真菌優化的培養基中。雖然以下培養條件涵蓋了大多數細菌和真菌,但一些需要特定條件的微生物在我們的測試中無法檢測到。所有用於分發的細胞系均已通過此測試,沒有受到污染。 中等的 含有 5% 兔血的血戰者 含 2% 酵母萃取物的營養肉湯(NB 培養基) 巰基乙酸鹽肉湯(TGC 培養基) 培養溫度:35℃ 培養期:1週-2個月(取決於培養基)
2.黴漿菌篩檢
黴漿菌污染是一個嚴重的問題,可能導致錯誤的數據並導致不必要的重複實驗。我們評估黴漿菌污染的常規程序包括以下三種方法。黴漿菌酵素活性 培養上清液的測量是檢查污染的快速測試。 試劑:MycoAlert™黴漿菌檢測試劑盒(Lonza, LT07-318) ☞ 使用 VERO 細胞進行螢光染色 將上清液樣本接種到 VERO 細胞培養物 (JCRB0111) 上。培養 5 天后,以 Hoechst 33258 對細胞進行染色,檢測 VERO 細胞的細胞核以及黴漿菌 DNA。如果黴漿菌呈陽性,則會在細胞核周圍觀察到額外的螢光,顯示蜘蛛網狀結構。透過這種方法也可以觀察細菌和真菌的污染。
3.巢式PCR
使用兩組引子進行支原體 DNA 擴增,從而提高了靈敏度和特異性。 (淨化) 當細胞被支原體污染時,以 MC-210 (DS Pharma Biomedical) 進行處理。處理後培養3個月確認去污。如果 MC-210 不起作用,則應用 Plasmocin (InvivoGen) 或 Plasmocure (InvivoGen)。
